i’ve to backup 1.5tb and i’ve a 4tb mycloud drive.
to save time, can i do the first backup with USB port and, after, do the incremental backup via wifi all with time machine?

No you can’t. The USB port is only to attach an external USB drive to the NAS. You could however connect the NAS directly to the computer using the ethernet port, then reboot the computer and the NAS, but this will not be faster than connecting both the NAS and the computer to your Gigabit switch in your router. That configuration, by the way, is faster than the USB connection at the back of this NAS.
