Do the new 16TB have two USB 3.1 or two USB 3.0?
They allow daisy chaining additional HDs.

Assuming two identical 16TB boxes daisy chained, would Win10 allow them to be treated as RAID 1?

My Book Duo have USB 3.0 speed. If you want to have a RAID 1 of 16TB then the drives have to be dynamic + they must be NTFS partitioned. Search online how to create a RAID 1 using external USB drives.
