Just cleaned up my Desktop drives and want to create a new clean back up. Can I or should I Format the Passport?

You don’t have to.  But if the drive contains any hidden files, like for Thumbnails, you can do a quick format to get rid of all that.  Just make sure that you have all your data backed up, or you don’t care to lose what’s on there.
