Is it true that the Samsung Smart TV has to be HARDWIRED in order to see the MyCloud Network drive?

I don’t know, but it seems unlikely.

What does its manual say?

Does it have built-in WiFi, or does it need a dongle?

Many manuals will recommend a hardwired connection because it is more reliable, since it doesn’t suffer interference or signal strength issues.

Can I use WIFI on my smart tv to access the MyCloud as long as both devices are on the same network?

Yes, you should be able to. I connect a number of devices to my MyCloud via WiFi on my local network, and stream media to them, using DLNA/UPnP apps.

If my Smart TV can natively play MKV files then in theory I should not have a problem playing them with MyCloud, correct?

In theory, yes…

Other users have reported that they have mixed results with Samsung smart TVs; old ones connect, new ones don’t. Compatibility with DLNA seems to be variable…
