You mean the internal hard drive in the my book? I would guess that is not possible. Try calling tech support to see if you can replace it. You can also check the link below for help.

Are you trying to replace the drive in the PC with the one from a My Book?

I have a similar question. The hard drive of the WD MyBook Essential 500GB is died (not a WD fault, it’s user error, if you understand ). Can I put a new hard drive into it? Of course, the MyBook Essential does have only one disk, no raid, etc. Or, can I use larger disk? For example a 2TB disk, instead of the original 500GB?
