No, it’s not a requirement – that just controlls the name that the DLNA clients will see for the server.

Well, all of those are “Containers,” and all (except VOB) can contain multiple media types.

Pure DVD ISOs are quite well supported, but not via DLNA (no DLNA server supports ISOs in any standard way). 

Most DLNA servers will support VOB files, but will not allow for DVD navigation (navigation is controlled via IFO files, not VOBs.)

Network Share-connected WDTV will support all of them, as long as the contents of the container are also supported (audio / video codec selection, etc.)
