If I buy one WD 2TB My Book Essential external drive, can I connect to one computer, back it up, then disconnect and connect to a second computer, and back that one up into separate folders, so that I have both computers backed up onto one external drive?
It depends what you are using for backup. I don’t think Smartware will work for that I’m not sure I don’t use it.

Yes you do not need to install smartware for that. If you want to backup Windows and Mac the best solution is to leave the drive formatted NTFS and go to Apple site and download and install NTFS-3G http://mac-free.com/download/NTFS-3G.html Then you will be able to use the drive with both Windows and Mac. Otherwis the drive will be read only on the Mac.
